#P2Solo @nanopore second trial of rapid PCR cDNA sequencing, running 18 samples on a PromethION P2 Solo flow cell.

[Our first trial was on a MinION flow cell]

I'm hoping to get over 25M reads from this run, which should be easily achievable given that it should get to 2M reads within the first hour, and it'll be running for 72 hours.

My current estimate is for about 70M reads by the end of the run.

in reply to David Eccles 🌻🇵🇸6x🩹🛡️

for my WGS work the pore occupancy drops off pretty quickly, so reloading may be more necessary, but yours seems pretty stable.
in reply to Frank Aylward

@foaylward long reads gel up the pores pretty quickly, which *is* a problem that we're having with this run, but it's not worth our time / money to re-use this flow cell for the same samples.

I'll probably wash it out and re-use it for lower-volume samples that I would have previously used a Flongle flow cell for.

Went to see a superblock in Barcelona. Given how much they feature as urbanism of the future, they are amazingly simple. Take an intersection, block traffic in all four directions and you have a square. Plant trees, add some tables and benches. That's basically it. Replace cars with trees and don't privatize the resulting space. All you need, and all that is usually missing, is political will.

Beautiful article!
The 72 Japanese micro-seasons

"Sandrine Bailly in her book Japan: Season by Season also references Murasaki Shikibu, the aristocratic observer in the Imperial court who authored The Tale of Genji as having written: “It is nature that gives me the most pleasure, the changes through the seasons, the blossoms and leaves of autumn and spring, the shifting patterns of the skies.”2

"Bailly concludes that much of Japanese art appreciates the evanescence and beauty of that which is to disappear. With clear influence from Buddhist thought, in Japan this beauty is often described by the term mono-no-aware—the poignant loveliness of the fragile universe. The Japanese imagination eulogizes that which is destined to fade and cherishes what is impermanent. It is in the very nature of fleetingness that we can feel the full impact of this delicate presence."

When I tell you that police are not designed to do what you think they're designed to do, you don't believe me.

81 kids, zip tied for hours, until some peed themselves.

While in other parts of the city, car break-ins happened.🤡

Later, you'll ask me why they don't trust cops.

New important article in Royal Society Open Science about the importance of academics to manage our own information networks:

"Demonstrating that the scholarly community is capable of creating a truly public square for scholarly discourse, impervious to private takeover, might renew confidence and inspire the community to focus on analogous solutions for the remaining scholarly record—encompassing text, data and code—to safeguard all publicly owned scholarly knowledge."

#AMD #EPYC #Bergamo is Fantastically Fresh Take on #CloudNative Compute
AMD was able to shrink each #CCD by adopting new #Zen4c core. AMD has less L3 cache (half at 2MB versus 4MB) with Zen 4c, and that allowed company to shrink area in a straightforward manner. Big change is transitioning to 12x 8-core CCDs on 96-core #Genoa to 8x 16-core CCD on Bergamo. Expect #EPYC9754 to be 15-20% faster than #EPYC9654 not a straight 33% as would be expected due to core count increase

History was made. I wasn't even born in that decade. 🚀 "Apollo 11 launched from Cape Kennedy on July 16, 1969, carrying Commander Neil Armstrong, Command Module Pilot Michael Collins and Lunar Module Pilot Edwin "Buzz" Aldrin into an initial Earth-orbit of 114 by 116 miles. An estimated 650 million people watched Armstrong's televised image and heard his voice describe the event as he took "...one small step for a man, one giant leap for mankind" on July 20, 1969." nasa.gov/mission_pages/apollo/…

any recommendations for specific values to use here? Like a levy that works well? Or is the point for us to experiment and work that out for ourselves? ;)

Looks interesting in any case, will try and convince my boardgames group to give this a try.
