Many in vitro #CRISPR knock-in approaches use small molecule drugs to improve the efficiency of homology-directed repair, but these drugs may or may not be safe in vivo. In this paper from #NatureBiotechnology, the authors find that an FDA-approved therapy, fludarabine, can be used for a similar purpose. Completely agree with this. I've long believed that our lives should be measured not by our grand accomplishments, but by the connections we make with other human beings, with nature, etc. Our lives should have an echo that lasts long after we're gone.

You've been on the ball lately, so my update process usually starts by hearing about some issue from you, and builds on whatever observations you make.

I agree it's a little odd how slow or irregular some organisations are in maintaining their news feeds. Seems there is an infection in parts of the FOSS space of the corporate mentality that saying literally anything constitutes a legally binding promise, and so some maintainers are afraid to keep their users in the loop.

Okay, so. My dorky #DevSecOps-learning ass wants to run Dependency-Track (dependencytrack.org/), but it specifically checks whether it can allocate 4GB of RAM on startup and dies if it can't. I've run out of pi 4s at home and yikes, I do not want to pay for a VPS with that much memory for playing with a single utility.

...so how can I fake out the Java/docker setup to get it to run under a different amount of memory? I don't care if it's slow. Can I... use disk swap or something? 🧐

Here's my (relatively) portable #famicom (#nes) #chiptune setup.

I can power it with a portable USB battery and it packs away easily into a small bag. Great way to get my #2A03, #MIDINES fix on the go!

There's so much micro-aggressions on here against black people. The thing with micro-aggressions is that it quickly piles up.

Mastodon culture, yes still on the "nice side", whatever that means, suffers from this still.

Any time you deny that there's anti-blackness on here, it's because you just aren't equipped to see it. Heck I'm not equipped to see it yet. So I'm going to defer to someone else.

if you didn't get it, the point is that being socko is just everyday for us.

most of the time we don't have our own voice because we'll not get listened to. the white people need to be willing to listen or to give us platforms and everytime they do if we don't choose carefully what we say we upset the white people and they may react violently in a way that is just plain accepted by everyone around us.

the angry black person stereotype is a testament of this. it is what happens when we're not willing to play by the rules white people set for us.

"oh please, explain to me this upsetting subject that I could go and learn myself" says the white people and when we get upset by the subject or by having to explain that to then they get violent.

do you have any fucking idea how taxing living like this is? how much energy goes into that?

I'd like to let you know that folks on the fediverse try to add alt text to their images. If you could delete and re-draft that would be great. I follow @PleaseCaption for reminders. I think it's a really good habit to get into that helps folks with screen readers access your content (and sometimes is a part of the humor) as well some folks (me included) won't boost without alt text.
